Cytoarchitectural comparism of the left ventricle and ascending aorta of antimalarial treated rats

Some of the effects of therapeutic, acute, and over dose treatment of Chloroquine, Artesunate, and Amodiaquine- Artesunate combined therapy on the lef t ventricle and ascending aorta of adult Wistar rats were studied using the routine Haematoxylin an d Eosin (H and E) and Orcein stains. The drugs were dissolved in distilled water and administered orally using an orogastric tube. The animals were sacrificed on days 3 and 5 after which the excised tissues were subjected to histological analysis. Appearance of autophagic vacuoles were evident in t he Chloroquine treated group and the sections revealed some varying degrees of disorientation and disarrangement of cardiac cells, with less stainin g intensity of elastic fibres occurring in the Orcein stained sections indicating breakdown of the cellu lar architecture of the left ventricle as a result of a cute overdose administration of the drugs with the most profound effect being evident in the Chloroquine tr eated group. It is concluded that acute overdose treatment of the drugs will have deleterious effect s on the metabolic activities of the left ventricle and ascending aorta of adult wistar rats.
